kinololResearchers at Avidin have synthesized analogues of novel quinoline analogues may have potential applications in anxiety disorders and Alzheimer’s disease (AD). The IP held by Avidin on the novel structure and its application is valuable, offering the possibility of widening the application of the drug candidate. Kinolol compounds exert neuroprotective action in vitro, they orally active as anti-anxiety drugs in vivo and have suspected activity against AD with minimal toxicity.

New Analogues

Avidin has synthesized more than 80 quinoline analogues and tested them in vitro for cytoprotective effects. 2 analogues have been already synthesized in kg quantities under GMP conditions with good impurity profile. We have preliminary data on stability, solubility and salt formation properties, as well as bioaviability. Kinolols are able to chelate ferric/zinc ions and have similar core structure as a drug candidate for AD which has positive results in a Phase IIb trial by Prana Biotechnology.

Pre-clinical

Results Avinomid showed impressive effectiveness of Q50 and Q134 kinolols in rats in elevated plus maze and forced swimming tests at 2-6 mg/kg oral dose. Q50 inhibited ischemic tissue damage in a heart transplantation rat model. Acute toxicology in mouse showed that kinolols are non-toxic up-to 60-120 mg/kg dose.

Applications

The main indication for which quinoline analogs are currently used is AD. Our new analogues showed impressive effectiveness against other indications (cytoprotection against oxidative damage, anxiety), however the application of kinolols would be wider than those currently suggested from cellular and animal studies. Overall market potential for anxiety disorders and AD in the world is app. 10$ Bn/year.

IP position

Avidin submitted a patent for the therapeutic application (cytoprotection, CNS applications) of kinolols, as new compounds. The patent was submitted in May, 2010, and PCT in May 2011, that enables long-term exploitation of the IP.

Financing

The preclinial development of Avinomid is currently financed by Avidin and Ubichem Ltds. Avidin is now seeking partners for licensing and co-development of kinolols to finish preclinical phase and to widen its applicability to Alzheimer’s disease.
